You can find almost any information about that board including many reference designs and ready to run code if you search a bit on the net. A good place to start your search would be TerasIC, so look at http://www.terasic.com.tw/cgi-bin/page/archive.pl?language=english&categoryno=65 and find the components you are working with. There you find all resources as PDF. Also download the VIP Suite User Guide from Altera and read about how to use the modules and what functions they have. (When you want to mix the two video streams you should take a closer look at the AlphaBlendingMixer.) I think you will first have to read a bit more into the details, perhaps if your knowledge about designing embedded systems is not so sophisticated even read a bit of the Quartus handbook or experiment with some easy example on the DE-2 first (like doing something with the LEDs).
