Can the INIT_DONE output pin be enabled for Cyclone V? If so, what is the process for enabling this feature?

Assignments -> Device -> Device and Pin Options...

Under the 'General' section, select 'Enable INIT_DONE output'

Yes. INIT_DONE output pin can be enabled in Cyclone V, purpose is to ensure successful configuration.
