Is there any way to test the ADA_THDB board without interfacing it without the cyclone 3 starter kit ? Am yet to order the starter kit and I want to verify the ADA board before the starter kit arrives.

Not really. The DAC needs data from somewhere and the ADC data needs to go somewhere. That "somewhere" is via the HSMC connector which is designed to interface to an Altera development kit. So you're pretty much stuck waiting for a development kit.
