At this modul how can I connect the output -tx_dataout[0] to the phisical output (output symbol)? Is this ouput a bus consisting of two lines (as it should be since the channel is diferential)? And i...

You only get the positive output. You connect it to the positive side of the differential pair in your design. You don't declare the negative side of the pair as a pin in your design. Quartus automatically infers the negative pin because of the IO standard used (PCML).
